Principais Tópicos de Pesquisa O estudo dos processos redox tem alto potencial para elucidar mecanismos biológicos

integradores, devido à prevalência desses processos e de seus fortes efeitos. Muitos estudos envolvem o

papel dos mecanismos redox em doenças. Contudo, a complexidade deste campo tem aumentado com o

conhecimento das estruturas biológicas, das relações estrutura-função, da patologia molecular, da

genômica, da proteômica e da metabolômica do estresse oxidativo. Assim, uma abordagem

multidisciplinar se tornou essencial para avanços significativos no campo. Nosso objetivo é realizar

estudos que permitam a concepção e avaliação de estratégias antioxidantes com aplicabilidade clínica.

Abordamos questões relevantes para superar as limitações no conhecimento dos processos redox, como:

1) Formação de ROS e controle em sistemas biológicos; 2) A reatividade química de espécies reativas de

oxigênio (ROS) em ambientes biológicos e consequentes mudanças na estrutura e função de

biomoléculas; 3) Mecanismos e redes envolvidas em processos de sinalização redox relevantes para

doenças humanas; 4) Diagnóstico e aplicações terapêuticas de processos redox.

Resumo dos Dados Obtidos e Perspectivas

O núcleo da rede Redoxoma começou no Programa Institutos do Milênio e adquiriu experiência

para fazer importantes contribuições para o avanço do conhecimento sobre as bases moleculares dos

processos redox no primeiro ano do INCT. É impossível resumir os resultados obtidos em poucas linhas,

mas aqueles que abrem novas perspectivas foram destaque em nosso Relatório Anual (ver Relatório

Anual em http://www2.iq.usp.br/redoxoma/). Com relação aos biomarcadores, foi desenvolvido um método

fluorescente, altamente sensível, para a detecção de aldeídos de colesterol formado por ozônio e oxigênio

singlete (Mansano et al., Anal Chem. 82:6775-81, 2010). Em relação às enzimas antioxidantes, novos

insights sobre a especificidade das interações tiorredoxina-tiorredoxina redutase foram fornecidos (Oliveira

et al., Biochemistry 49:3317-26, 2010). Demonstrou-se que a geração de espécies reativas de oxigênio

pelas mitocôndrias é substrato-, tecido- e sítio-específica (Tahara et al., Free Radic Biol Med 46:1283-97,

2010). Com relação aos danos a biomoléculas, foi demonstrado que a proteína de recombinação RAD52

coopera com a proteína OGG1 para reparar lesões oxidativas no DNA de células de mamíferos (de Souza-

Pinto et al., Mol Cell Biol 29:4441-54, 2009). A elucidação de mecanismos pelos quais a dissulfeto

isomerase protéica (PDI) regula processos redox via interação com NADPH oxidase vem revelando vias

de sinalização importantes e passíveis de intervenção terapêutica em vários modelos de doença

cardiovascular (Santos et al., Antioxid Redox Signal. 11:2409-27, 2009). Também foi definida uma rede de

proteínas em Saccharomyces cerevisiae relacionada ao processo de envelhecimento (Barea e Bonatto,

Mech Ag Devel 13:444-60, 2009). Aplicações terapêuticas de processos redox foram exploradas com o

antioxidante não clássico tempol, que se mostrou eficaz no tratamento da esclerose múltipla em

camundongos (Tsuhako et al., Free Radic Biol Med 48:704-12, 2010).

Comentários gerais

Os grupos que compõem a rede Redoxoma produziram 194 artigos científicos, 14 capítulos de

livros e editaram um livro, no período de 2009-2010. Algumas das nossas contribuições científicas foram

publicadas em conceituadas revistas gerais, tais como FASEB J, J Biol Chem, J Mol Biol e Biochemistry;

outras contribuições foram publicadas em revistas especializadas de alto impacto, tais como Ant Redox

Sig e Free Rad Biol Med. A maioria dessas contribuições é consequência da qualidade dos vários grupos

que compõem o Redoxoma. A influência da rede em si é ainda recente, mas a interação entre os vários

grupos está se expandindo, como atestam os vários trabalhos publicados em conjunto (Fig. 1). A lista

completa das publicações de 2009 e 2010 é apresentada abaixo, com as publicações em revistas de

impacto ≥ 5 em azul. Na área de Bioquímica e Biologia Molecular, 18,7% dos periódicos têm esse índice

de impacto, de acordo com o Journal Citation Reports ISI Web of Knowledge (HTTP://admin-

apps.isiknowledge.com/JCR/JCR).
