RESUMO
Ao longo das dcadas, a quantidade de informaes sobre recursos medicinais a partir do uso
de plantas das florestas tropicais aumentou significantemente. A regio amaznica do Brasil
devido sua grande diversidade biolgica de relevante importncia para descoberta de
substncias com potencial medicinal e principalmente em relao a este estudo, a atividade
antibacteriana. As drogas antifngicas sistmicas so em nmero bastante limitado e, alm
disso, apresentam efeitos colaterais que podem limitar sua utilizao, portanto importante a
procura de novas drogas antifngicas que apresentem maior segurana na sua utilizao e
menos efeitos colaterais. Dentre a bioproduo oriunda da Amaznia destaca-se a famlia
Piperaceae com distribuio principalmente pantropical que compreende 12 gneros e 1400
espcies. O gnero Piper o maior da famlia, com 700 espcies. Entre elas, h 170 espcies
que ocorrem no Brasil sendo utilizadas popularmente como renomados agente anti-sptico,
inseticida e antimicrobiano. O objetivo desse trabalho foi elucidar os componentes qumicos e
a atividade dos leos essenciais de espcies de Piper da Amaznia Brasileira contra cepas de
Candida spp. Folhas das espcies Piper marginatum, P. aleyreanum, P. dilatatum, P.
aduncum e P. alatabaccum foram coletados na floresta amaznica para a avaliao da
atividade antifngica contra nove cepas de Candida spp., incluindo trs cepas isoladas de
sangue com MICs elevados para o fluconazol. As cepas foram identificadas em
CHROMagar. Os leos essenciais foram obtidos por hidrodestilao e testados na faixa de 10
a 0,015 mg/mL pelo mtodo de microdiluio em caldo de acordo com o protocolo M27-A3.
As cepas de C. parapsilosis ATCC 22019 e ATCC 6258 foram utilizadas como controle. Os
principais componentes dos leos essenciais foram para P. marginatum foram: -copaeno e
3,4-(metilenodioxi) propiofenona, para P. aleyreanum: E-cariofileno e xido de cariofileno,
para P. dilatatum: -copaeno, xido de cariofileno, linoleato de metila e octadecanoato de
metila, por P. alatabaccum: E-cariofileno, spathulenol, metil linoleato e metil octadecanoate e
para P. aduncum: trans-isocroweacin, dill apiol e cis-isoelemicin. P. dilatatum foi o mais
ativo com MIC50 de 0,65 0,39 mg / mL. Os outros leos mostraram MIC50 variando de 1,55
0,77-2,10 0,75 mg / mL, confirmando a importncia dos leos essenciais de Piper como
alternativas naturais para o tratamento de infeces causadas por Candida spp.
ABSTRACT
Over the decades, the amount of resource information from the medicinal use of plants of
tropical forests has increased significantly. The Amazon region of Brazil, due to its great
biological diversity, is of great significance for the discovery of substances with medicinal
potential. The systemic antifungal drugs are very limited in number and, moreover, have side
effects that may limit its use, therefore it is important the search for new antifungal drugs safe
for the utilization. Among bioproduction coming from the Amazon, stands out the Piperaceae
family, with pantropical distribution, primarily comprising twelve genera and 1400 species.
The genus Piper is the largest family with 700 species. Among them there are 170 species
occurring in Brazil are popularly used as renowned antiseptic agent, insecticide and
antimicrobial. The aim of this study was to determine the chemical components and activity
of essential oils from Piper species of the Brazilian Amazon against strains of Candida spp.
Leaves of the species Piper marginatum, P. aleyreanum, P. dilatatum, P. aduncum and
P. alatabaccum were collected in the Amazon forest for evaluation of antifungal activity
against nine Candida spp. strains, including three strains isolated from blood and being
fluconazole-resistant. The strains were identified on CHROMagar. The essential oils were
obtained by hydrodistillation of plant leaves and tested in the range of 10 to 0.015 mg/mL by
broth microdilution method according to protocol M27-A3. The strains C. parapsilosis ATCC
22019 and ATCC 6258 were used as controls. The main components of the plant essential oils
were for P. marginatum: -copaene and 3,4-(methylenedioxy) propiophenone, for
P. aleyreanum: E-caryophyllene, caryophyllene oxide, for P. dilatatum: -copaene,
caryophyllene oxide, methyl linoleate, methyl octadecanoate, for P. alatabaccum: E-
caryophyllene, spathulenol, methyl linoleate, methyl octadecanoate and for P. aduncum:
trans-isocrowecin, dill apiol and cis-isoelemicin. P. dilatatum was the most active with MIC50
of 0.65 0.39 mg/mL and other showed MIC50 ranging from 1.55 0.77 to 2.10 0.75
mg/mL, demonstrating the importance of Piper essential oils as natural alternatives for the
treatment of infections caused by Candida spp.

1 INTRODUO
A procura por novos agentes antimicrobianos, a partir de plantas, intensa devido
resistncia dos microorganismos patognicos frente aos produtos sintticos (BAUTISTA-
BAOS et al., 2003). A resistncia aos antifngicos, clnica ou in vitro, pode ocorrer quando
clulas de fungos suscetveis passam a ser resistentes devido ao contato prvio com as drogas
ou inerente ao prprio microrganismo. (MORSCHHUSER, 2002)
Os produtos naturais tm sido muito utilizados como modelos para o
desenvolvimento de novos compostos antimicrobianos e os resultados apresentam grande
utilidade contra doenas fngicas (DENNING, 2002). O Brasil o pas com maior
diversidade gentica vegetal do mundo, contando com mais de 55.000 espcies catalogadas
(AZEVEDO & SILVA, 2006), frente a esta diversidade uma estratgia atual vem sendo
buscar mtodos alternativos para o controle de doenas e pestes, que visem causar menos
danos ao ambiente e a sade humana. (MOREIRA et al., 2004). Dentre os agentes
teraputicos provenientes das plantas com uso medicinal popular e cientfico, destacam-se os
leos essenciais. (COSTA et al., 2008)
Trabalhos desenvolvidos com leos essenciais, obtidos a partir de plantas
medicinais tm indicado atividade no controle de fitopatgenos, substituindo o uso de
pesticidas que em longo prazo causam impactos negativos para a sociedade e para o meio
ambiente devido poluio causada pelos resduos qumicos dos mesmos (MOREIRA et al.,
2004). Alguns exemplos so o leo essencial extrado de Caesulia axillaris com desempenho
superior a oito fungicidas sintticos e eficcia no controle da mancha foliar do arroz, causada
por Helminthosporium oryzae (AMADIOHA, 2000) e o leo de Piper aduncum com
comprovada ao inibitria contra vrios fitopatgenos destre eles Crinipellis perniciosa,
Phytophthora palmivora e Phytophthora capsici. (BASTOS & SILVA, 2002)
Muitas das espcies de Piper so usadas para fins curativos por diversas culturas,
tendo a sua constituio sido composta por amidas insaturadas, flavonides, lignanas,
aristolactamas, steres, de cadeias longas e curtas, terpenos, esterides, propenilfenois e
alcalides (PARMAR et al., 1997). O leo essencial das folhas de P. hispidum possui como
principais componentes -canfeno guaiene e -elemeno (FACUNDO et al., 1994). As folhas
de P. amalago so utilizadas no Mxico e no Brasil para aliviar dores estomacais e no
combate a diversas infeces. No estado da Paraba folhas e talos de P. marginatum e P.

tuberculatum so utilizados contra picada de cobra e como sedativos (GUIMARES &
GIORDANO, 2004). Na China, algumas prescries recomendam o uso das folhas de Piper
futokasura no tratamento de arritmias cardacas e da asma. Infuses das folhas de P. aduncum
e P. hispidum so utilizadas na Jamaica para o tratamento de dores estomacais. (PARMAR et
al. 1997). A espcie Piper aleyreanum, muito usada por populaes tradicionais e indgenas
da regio Amaznica, devido suas propriedades anestsica no alvio de dores (JOLY, 1998).
Piper carniconnectivum uma espcie encontrada na Amaznia, na regio norte do Brasil
sendo til como bioprodutora de leo essencial. (RIBEIRO et al, 1999)
A ao antimicrobiana dos leos essenciais, tanto in vitro como in vivo, tem
justificado a investigao sobre a medicina tradicional centrada na caracterizao de sua
atividade antimicrobiana. (AGARWAL et al., 2010)
Os antifngicos podem ser agrupados em trs classes baseados no seu ponto de
ao, inibidores da sntese do ergosterol (o principal esterol de fungos); polienos, que
interagem fisicoquimicamente com os esteris de membrana e inibidores da sntese de
protena (ARTHINGTON-SKAGGS et al., 1999). leos essenciais so relatados como
inibidores da sntese de ergosterol. (BARBOSA, 2010)
Neste trabalho foi avaliada a atividade antifngica dos leos essenciais P.
marginatum Jacq, P. aleyreanum C.DC., P. dilatatum L. C. Rich., P. aduncum L. e P.
alatabaccum Trel & Yuncker contra cepas de Candida spp, isoladas no Cear.

2.1 PLANTAS MEDICINAIS E O GNERO PIPER: BREVE HISTRICO E
PERSPECTIVAS
Iniciada antes da medicina moderna, a medicina tradicional definida pela
Organizao Mundial da Sade (OMS) como sendo a soma de todos os conhecimentos
tericos e prticos, explicveis ou no, utilizados para diagnstico, preveno e tratamentos
fsicos, mentais ou sociais, baseados exclusivamente na experincia e observao e
transmitidos verbalmente ou por escrito de uma gerao a outra. (PEREIRA, OLIVEIRA,
LEMOS, 2004)
As plantas medicinais tm sido utilizadas por milhares de anos como tratamento
alternativo para a cura de diversos males. A maioria da populao dos pases em
desenvolvimento ainda utiliza as plantas medicinais como principal fonte para suprir as suas
necessidades medicamentosas. Mesmo em pases industrializados, uma grande percentagem
dos produtos farmacuticos comercializados provm de produtos naturais e essa proporo
eleva-se em regies do mundo, como sia, frica e Amrica Latina, onde vive a maior parte
da populao mundial. (PIMENTA, 2008)
Em 1913 a Flora Mdica Brasiliense, ressalta a importncia de serem estudadas,
metdica e cientificamente, as plantas da Amaznia num trabalho conjunto de botnicos,
qumicos, bilogos, farmaclogos e clnicos. (MATTA, 1913)
No entanto, a atividade antimicrobiana das plantas comeou a ser estudada
somente na dcada de 1940 quando Osborn (1943), pesquisando a atividade, in vitro, de 2.300
plantas superiores, contra Staphylococcus aureus e Escherichia coli, verificou que 63 gneros
continham substncias que inibiram o crescimento de um ou de ambos os microrganismos.
(SARTORI, 2005)
Bastos (1997) justificou que o leo essencial das folhas de P. aduncum foi efetivo
sobre a germinao de basidisporos e crescimento micelial de Crinipellis perniciosa.
Atualmente as espcies de Piper so classificadas como grandes produtoras de leos
essenciais. (SILVA & BASTOS 2007)
A famlia Piperaceae classificada entre as mais primitivas famlias entre as
angiospermas e compreende aproximadamente trs mil espcies distribudas em oito gneros
(DI STASI et al.,2002), dos quais se destacam os gneros Piper, Peperomia e Pothormorphe.

O gnero Piper possui por volta de 700 espcies distribudas em todas as regies tropicais do
mundo. (SILVA & BASTOS, 2007)
No Brasil, ocorrem cerca de 266 espcies do gnero Piper, das quais 13 espcies e
4 variedades foram descritas apenas no estado do Cear, embora praticamente todas tambm
ocorram em outros estados nacionais. Da Amaznia, vrias espcies j foram estudadas como
P. belte, P. nigrum, P. amapaense, P. duckei, P. bartlingianum e P. arboreum (GUIMARES
& GIORDANO, 2004). O quadro 1 sumariza a ocorrncia e as principais atividades
medicinais das diversas espcies do gnero Piper nos estados brasileiros. A maioria das
espcies deste gnero no tem valor econmico, embora apresentem uma grande importncia
ecolgica dentro do seu habitat natural, principalmente nas florestas midas. As folhas das
plantas do gnero Piper servem de alimento para uma srie de insetos herbvoros, enquanto
que os frutos amadurecidos nutrem uma variedade de animais frugvoros, por exemplo,
morcegos e pssaros (DYER et al., 2004). No entanto, algumas plantas do gnero Piper so
economicamente muito importantes. Um exemplo a espcie P. methysticum cujas razes so
fontes de kawa, substncia da classe das kavapironas, usada tradicionalmente na Polinsia
como uma bebida narctica, hoje, esta planta de interesse para as indstrias na formulao
de sedativos. Outras espcies complacentes como P. cubeba, P. longum e P. auritum so
usadas localmente como condimentos ou medicinais. (DYER et al., 2004).
Em trabalho realizado com treze plantas utilizadas na medicina popular para o
tratamento de doenas infecciosas P. regnellii apresentou atividade frente a Candida krusei e
C. tropicalis.(PESSINI, et al., 2003). Segundo Silva e Bastos (2007) leos essenciais de P.
callosum, P. marginatum var. anisatum, P. callosum e P. ebnckea tambm apresentaram
excelente atividade frente C. perniciosa.
Os leos essenciais constituem os elementos volteis contidos em muitos rgos
vegetais e esto relacionados com funes necessrias sobrevivncia vegetal, exercendo
papel fundamental na defesa contra microrganismos. (SIQUI et al., 2000)
Praticamente todos os leos essenciais constituem uma mistura de princpios
qumicos, muito complexa e variam amplamente em sua composio, sendo a maior parte
constituda principalmente de terpenos (MATOS et al., 1989). Devido a sua composio
variada, os leos essenciais demonstram uma variedade de aes farmacolgicas, tornando-os
potenciais fontes para o desenvolvimento de novas drogas. (AMARAL, 2004)
Nos ltimos anos a atividade antifngica de leos essenciais tem sido relatada
para diversas espcies, envolvendo principalmente as famlias Lamiaceae, Asteraceae,
Verbenaceae, Rutaceae, Lauraceae e Cupressaceae (ABAD et al., 2007). Na regio

Amaznica, tem-se uma das maiores biodiversidades, com um nmero satisfatrio de leos
essenciais e extratos utilizados, popularmente, no tratamento de infeces fngicas. Dentre as
diversas espcies merecem destaque Piper aduncum (pimenta-de-macaco) e Piper
hispidinervum (pimenta longa) (MENEZES, et al., 2009).
Neste trabalho foram utilizados os leos essenciais de Piper spp. para
determinao de sua atividade antifngica.

2.2.1 P. marginatum Jacq

P. marginatum conhecida popularmente como caapeba cheirosa ou malvarisco.
As folhas so utilizadas na forma de infuso da raiz e da folha para o tratamento de dores em
geral, febre, gases, blenorragia, afeces do fgado e antdoto contra picada de cobra. Seus
frutos so usados como substitutos da pimenta-do-reino, utilizando-os como condimento
(ALMEIDA, 2008). J foram isolados das razes de P. marginatum l, 2-metoxi-4,5-
metilenodioxipropifenol e (E,E)-N-isobutil-2,4-octadienamida. A partir do extrato hexanico
dessa espcie foram isolados: 1-(1-Z-propenil)-2,4,6-trimetoxibenzeno e cido 3-farnesil-4-
hidroxibenzoico e do extrato clorofrmico foi isolado xido cariofileno (fig.2) (CHAVES,
2002). Em um recente estudo de populaes de P. marginatum de vrios ecossistemas da
Amaznia brasileira, (ANDRADE., et al. 2008) foi descrito sete quimiotipos com base nas
diferenas da composio do leo das folhas.

2.2.2 P. aleyreanum C.DC
A espcie P. aleyreanum conhecida pelas populaes indgenas como pani
nixpu sendo utilizada para a sade dos dentes; os Kaxinaw dizem que seu sumo os
fortalece e protege (MCCALLUM, 1996). Em estudo realizado sobre a composio qumica
das folhas dessa espcie por FACUNDO & MORAIS (2003) foram relatados para a mesma
como componentes majoritrios galangina, 2- metoxi-4,5-metilenodioxipropifenol e
sitosterol, sendo este o primeiro relato da ocorrncia de galangina no gnero Piper; Galangina
foi previamenteisolado de brotos de Populus nigra (EGGER et al., 1969) e de folhas de
Castanea sativa (BASILE et al., 2000). -pineno, isocaryophylleno (fig.3) e -cariofileno
tambm j foram encontrados na composio qumica do leo essencial de P. aleyreanum
(FACUNDO, 2007). No foram encontrados outros relatos na literatura para P. aleyreanum

2.2.3 P. dilatatum Rich
Piper dilatatum um arbusto de 1-2 m de altura, conhecida popularmente como
pariparoba-murta e pau-de-junta suas folhas tm ao antiinflamatria e cicatrizante,
aliviando crises de bronquite e asma, lceras, reumatismos e hemorridas, e suas razes so
usadas no tratamento de odontalgias. Em estudo comparando a atividade fungitxica de leo

essencial de 10 espcies de Piper coletadas na regio Amaznica, sobre o crescimento
micelial e a germinao de basidisporos de Crinipellis perniciosa e sobre o crescimento
micelial de Phytophthora palmivora e P. capsici o leo de P. dilatatum foi o mais eficiente.
Na composio do leo essencial de P.dilatatum j foi relatada a presena de cis--ocimeno,
-cariofileno, germacreno D, espatulenol (fig.4) e xido de cariofileno (fig.2) (BASTOS
2007, SANTOS, 2001).

2.2.4 P. aduncum
um arbusto de ampla distribuio tropical, com ocorrncia em solos areno-
argilosos, conhecido popularmente como pimenta-de-macaco e aperta-ruo. Esta espcie
considerada uma planta oportunista que invade reas desflorestadas aps explorao de
madeira, de alta rusticidade e elevada resistncia s mudanas climticas. O uso medicinal de
pimenta-de-macaco tem sido relatado: em doenas ginecolgicas e desordens intestinais
(SOUZA, 2008), como diurtico, antiblenorrgico, carminativo, excitante digestivo, para
males do fgado, no combate a erisipela e tratamento de ulceras crnicas (COIMBRA, 1994).
Extratos orgnicos das folhas de P. aduncum apresentaram atividades moluscicida, citotxica
e antibacteriana, para as quais se associou a presena de dihidrochalconas e derivados
prenilados do cido benzico (ORJALA et al., 1993 & 1994).

O leo essencial de P. aduncum foi testado contra o fungo Clinipellis perniciosa,
conhecido como vassoura-de-bruxa, responsvel por ataque patognico ao cacau e cupuau.
Na concentrao de 50 a 100 ppm inibiu 100% o crescimento e a germinao deste fungo
(BASTOS, 1997). O leo essencial de P. aduncum apresentou atividade inseticida e larvicida
contra insetos fitfagos e mosquitos transmissores de dengue e malria, eliminando-os

totalmente em baixas concentraes (SOUTO, 2006). Em sua composio qumica esto
Dilapiol, piperitona, terpinen-4-ol e -terpineno. (MAIA, 2000)
2.2.5 P. alatabaccum Trel & Yuncker
P. alatabaccum Trel & Yuncker (Piperaceae), conhecida pelo nome vernculo
''Joo brandinho'', uma espcie que ocorre na regio amaznica no norte do Brasil
(RIBEIRO et al., 1999). Na composio dessa espcie esto N-(3,4,5-
trimethoxydihydrocinnamoyl-D3-pyridin-2-one, Piplartine, N-(3,4,5-trimethoxycinnamoyl-
D3- pyridin-2-one, Piperovatine (N-isobutil-6-p-methoxyphenylsorbamide) e 5,5,7-
trimethoxy-3,4-methylenedioxyflavone. Este o primeiro relato de 5,5,7-Trimethoxy-3,4-
methylenedioxyflavone no gnero Piper (FACUNDO et al., 2003). No foram encontrados
na literatura outros relatos para P. alatabaccum.
2.3 FUNGOS E SUA IMPORTNCIA NA MEDICINA VETERINRIA
Os fungos apresentam semelhana s clulas hospedeiras, sendo assim, a
elaborao de estratgias teraputicas especficas dirigidas contra o parasita e atxicas para o
hospedeiro se tornam difceis. Os mesmos so eucariticos, com ncleo bem definido
circundado por uma membrana nuclear; uma membrana celular que contm lipdeos,
glicoprotenas e esteris; parede celular; mitocndrias; aparelho de Golgi; ribossomos ligados
ao retculo endoplasmtico e um citoesqueleto constitudo por microtbulos, microfilamentos
e filamentos intermedirios. (SCHAECHTER et al., 2002). De acordo com sua morfologia os
fungos podem ser classificados em filamentosos, leveduras e dimrficos. (SIDRIM &
ROCHA, 2004)
As micoses podem ser classificadas, segundo os tecidos e rgos atingidos em:
micoses superficiais, de localizao nas camadas mais superficiais da pele ou dos plos;
micoses cutneas ou dermatomicoses, localizadas na pele, no plo ou nas unhas; micoses
subcutneas; micoses sistmicas ou profundas atingindo, principalmente, rgos internos e
vsceras, e as micoses oportunistas que atingem os pacientes imunocomprometidos por
doena de base, como cncer, diabetes, ou aqueles que so submetidos a tratamentos com uso
de corticoidoterapia, imunossupressores e antibioticoterapia. (TRABULSI et al., 2004)

As dermatofitoses so micoses superficiais que se localizam nas zonas planas e
intertriginosas da pele (LACAZ et al., 2002), e so produzidas por fungos especializados,
denominados dermatfitos, os quais tm a habilidade de degradar a queratina e transform-la
em material nutritivo para seu crescimento (SIDRIM & ROCHA, 2004). No Cear, as
espcies mais envolvidas em dermatofitoses so o T. rubrum seguido pelo T. tonsurans, M.
canis, T. mentagrophytes e E. floccosum. (BRILHANTE et al., 2000)
As enfermidades micticas tm importncia distinta dentro das enfermidades
veterinrias devido ao seu potencial zoontico (NAKAMURA et al., 1999).O sucesso no
tratamento pode estar relacionado ao conhecimento do patgeno, e por conseqncia, do seu
perfil ecolgico. (VIEIRA, 2009)
Fungos leveduriformes possuem importncia veterinria, como espcies dos
gneros Candida e Malassezia. O gnero Candida composto por fungos leveduriformes
hialinos, que apresentam duas formas de reproduo: assexuada ou anamorfa e sexuada ou
teleomorfa. Taxonomicamente so enquadradas dentro do grupo dos ascomicetos, visto que, a
reproduo sexuada caracterizada pela produo de ascos. (SIDRIM & ROCHA, 2004)
As espcies de Candida apresentam colnias de colorao branca, superfcie lisa
ou levemente rugosa e textura glabrosa mida (MORETTI et al., 2004), que crescem bem
dentro de 48 horas, entre temperaturas de 25 e 37C (SIDRIM & ROCHA, 2004).

Com relao microscopia, lminas preparadas diretamente a partir de um
fragmento da colnia ou de uma amostra clnica positiva, permitem a visualizao apenas de
estruturas unicelulares, denominadas blastocondeos, que podem estar isoladas ou

apresentando brotamento. Desta forma, torna-se necessrio a realizao de microcultivo em
placa de Petri, para melhor visualizao da disposio das estruturas fngicas tpicas de cada
espcie, sendo este, na maioria das vezes, suficiente para identificar o microrganismo.

Candida albicans causa 5060% das todas as candidemias diagnosticadas seguida
de C. parapsilosis (10-20%) e C. tropicalis (6-7%). Infeces da corrente sangunea por
Candida spp. apresentam uma mortalidade elevada e so responsveis pelo aumento dos
custos hospitalares. (ALFONSO et al., 2010)
Infeces em animais causadas por Candida spp. so freqentes (MULLER et al.,
2002) e no Brasil a presena de C. albicans foi detectada em bovinos com otite (DUARTE et
al., 2001) e em casos de dermatomicoses em ces (RAPOSO et al., 1996). Alm de C.
albicans, as espcies mais prevalentes so C. parapsilosis e C. guilliermondii, tendo sido
relatadas como responsveis por dermatomicoses em ces. (MULLER et al., 2002)
Candida spp. um habitante normal do trato gastrintestinal das aves, sendo a
espcie mais comumente isolada nesses animais. Entretanto o desequilbrio populacional
dessas leveduras leva ao aparecimento de problemas no sistema digestivo, podendo infectar
tambm o sistema reprodutivo, olhos e tornar-se sistmica. (GODOY, 2001).
A candidase em aves apresenta como sintomatologia dispnia, anorexia,
prostrao, regurgitao, vmito, diarria, perda de peso, inglvio dilatado, esofagite e

espessamento da parede do esfago. Casos raros e graves podem provocar deformidades no
bico. (BRAGA et al., 2007).
A histria natural das doenas causadas por leveduras mais bem compreendida
luz da susceptibilidade do hospedeiro. O indivduo normal apresenta mecanismos de defesa
inespecficos e especficos, tais como barreiras anatmicas e fisiolgicas, resposta
inflamatria e resposta imunolgica, que, juntos, representam obstculo ao estabelecimento da
infeco fngica. (VIEIRA, 2009)
2.4 MTODOS DE ESTUDO DA ATIVIDADE ANTIFNGICA IN VITRO
A fim de determinar a potncia de um antibitico em soluo, sua concentrao
em lquidos e tecidos corporais e a sensibilidade de um determinado microrganismo a
concentraes conhecidas deste antibitico a atividade antifngica medida in vitro.
(JAWETZ et al., 2005)
O Clinical Latoratory Standards Institute (CLSI, anteriormente denominado
National Committee for Clinical Laboratory Standards - NCCLS) nos Estados Unidos a
entidade responsvel pela normatizao de tcnicas de laboratrio clnico que padronizou os
testes de sensibilidade de fungos para drogas antimicticas, definindo variveis como mtodo
e preparao de inculo, composio e pH do meio a ser utilizado, temperatura e tempo de
incubao e determinao dos critrios de leitura do teste. (VIEIRA, 2009)
O primeiro mtodo padronizado pelo CLSI (1992) foi a macrodiluio em caldo,
porm, por tratar-se de mtodo laborioso e de difcil execuo em laboratrios de rotina,
houve a busca de mtodos alternativos. Entre os mais estudados, citam-se a microdiluio em
caldo com bons resultados e, mais recentemente, o Etest.

A microdiluio em placa, realizada segundo parmetros do CLSI, alm da maior
facilidade na sua execuo, permite a anlise de grande nmero de amostras, com economia
de material. (SIDRIM & ROCHA, 2004)
Outros procedimentos, tais como o mtodo de difuso em cavidade gar, apesar
de no ser padronizado pelo CLSI, possibilita anlises qualitativas, permitindo-se fazer uma
comparao da eficincia da droga testada com outra droga utilizada como referncia. Estudos
posteriores so necessrios para determinao da concentrao inibitria mnima (MIC) in
vitro, para que seja possvel correlacionar com resultados in vivo.

3 JUSTIFICATIVA
O aumento da incidncia de infeces causadas por fungos nas clinicas veterinrias,
juntamente com a resistncia que estes agentes tm desenvolvido aos antimicrobianos, tem
levado a uma busca constante por alternativas teraputicas eficazes que possam oferecer
melhores opes de tratamento. No Norte e Nordeste do Brasil varias plantas so utilizadas
pela populao para o tratamento de diversas enfermidades, principalmente em locais mais
carentes. Para o seu uso em larga escala, no entanto, estudos que comprovem essas atividades
fazem-se necessrios. Deste modo, um estudo detalhado a cerca da atividade antifngica dos
leos essenciais de espcies do gnero Piper entre essas, P. marginatum Jacq, P. aleyreanum
C.DC., P. dilatatum L. C. Rich., P. aduncum L. e P. alatabaccum Trel & Yuncker
importante para sua validao como provveis substncias com atividade contra fungos
frequentemente isolados de animais em clinicas veterinrias.

5 OBJETIVOS
5.1 OBJETIVO GERAL
Determinar a constituio qumica e avaliar a atividade antifngica in vitro dos
leos essenciais de Piper da Amaznia Brasileira contra cepas de Candida spp, isoladas no
Cear.
5.2 OBJETIVOS ESPECFICOS
Determinar a composio qumica dos leos essenciais de P. marginatum, P.
aleyreanum, P. dilatatum, P. aduncum e P. alatabaccum por cromatografia gasosa
acoplada a espectrometria de massa.
Avaliar a atividade antifngica dos leos essenciais de P. marginatum, P. aleyreanum,
P. dilatatum, P. aduncum e P. alatabaccum pelo mtodo de microdiluio em caldo
contra fungos do gnero Candida.

Resumo
O gnero Piper tem 170 espcies no Brasil, a maioria contem terpenos e compostos fenlicos
com atividade antimicrobiana. Folhas das espcies Piper marginatum, P. aleyreanum, P.
dilatatum, P. aduncum e P. alatabaccum foram coletados na floresta amaznica para a
avaliao da atividade antifngica contra nove cepas de Candida spp., incluindo trs cepas
isoladas do sangue com MICs elevados para o fluconazol. As cepas foram identificadas em
CHROMagar. Os leos essenciais foram testados na faixa de 10 a 0,015 mg/mL pelo mtodo
de microdiluio em caldo de acordo com o protocolo M27-A3. As cepas de C. parapsilosis
ATCC 22019 e ATCC 6258 foram utilizadas como controle. Os principais componentes para
P. marginatum foram: -copaeno e 3,4-(methylenedioxy) propiophenone, para P.
aleyreanum: E-cariofileno e xido de cariofileno, para P. dilatatum: -copaeno, xido de
cariofileno, metil linoleato e metil octadecanoato, por P. alatabaccum: E-cariofileno,
spathulenol, metil linoleato e metil octadecanoate e para P. aduncum: trans-isocrowecin, dill
apiol e cis-isoelemicin. P. dilatatum foi o mais ativo com MIC50 de 0,65 0,39 mg / mL. Os
outros leos mostraram MIC50 variando de 1,55 0,77-2,10 0,75 mg / mL, demonstrando a
importncia dos leos essenciais de Piper como alternativas naturais para o tratamento de
infeces causadas por Candida spp.

1. Introduction
The Piperaceae family out stands among the biodiversity of the Amazon forest with
mainly a pantropical distribution, comprising twelve genera and 1400 species. The genus
Piper is the largest Family with 700 species. Among them are 170 species, which occur in
Brazil. Most species are fast growing and popularly used as antiseptic, insecticidal and
antimicrobial agents (Fazolin et al., 2007, Duarte et al., 2007). In the Amazon Rainforest,
many of the native tribes use P. aduncum (matico) leaves as an antiseptic and wound healing.
In Peru, it was used for stopping hemorrhages and treating ulcers, and in European practice in
the treatment of diseases of the genitals and urinary organs and for treatment of haemostasis
and dysentery in Colombian ethnobotany (Rali et al., 2007). Piper marginatum is reputed in
the Brazilian folk medicine for its analgesic / antiinflammatory, hemostatic and skin wound-
healing properties (DAngelo et al., 1997).
Chemical studies showed that the genus Piper has many components, including
unsaturated amides, flavonoids, lignans, aristolactams, long and short chain esters, essential
oils, steroids and alkaloids (Facundo et al., 2005). The antimicrobial activity of essential
oils, both in vitro and in vivo, has justified the research on these plant components for
microbial infections (Agarwal et al., 2010)
Candidiasis is present in humans and animals and can affect mucous membranes and
mucocutaneous junctions or places where humidity is persistent. It causes papular, pustular
dermatitis or ulcerative and as well exudative external otitis. Candida albicans is yeast found
in mucocutaneous regions, genital and digestive tract of mammals and birds, being an
opportunistic pathogen producing more frequently stomatitis and vaginitis, but it could reach
the skin, fingernails, respiratory tract and lead to septicemia, depending on the degree of
immunosuppression of the individual. In dogs, candidiasis can also present as septicemia with

lesions in the muscles, bones and skin and is caused by various species of Candida (Raposo et
al., 1996). Candida albicans causes 50-60% of all diagnosed candidemia followed by
C. parapsilosis (10-20%) and C. tropicalis (6.7%). Bloodstream infections by Candida spp.
have a high mortality and are responsible for increasing hospital costs (Alfonso et al., 2010).
The systemic antifungal drugs are very limited in number and, moreover, have side effects
that may difficult its use, then, the discovery of new antifungal drugs should be pursued
continuously (Dolan et al., 2009).
Essential oil from plants are targets for achieving this goal, therefore this study was
designed to evaluate the antifungal activity of essential oils from Piper species of the
Brazilian Amazon forest against strains of Candida spp to justify its popular use in microbial
infection diseases.
2. Materials and methods
2.1. Plant materials and isolation of essential oil
The leaves of the species P. marginatum Jacq, P. aleyreanum C.DC., P. dilatatum L. C.
Rich., P. aduncum L. and P. alatabaccum Trel & Yuncker were collected between nine and
eleven hours of january 2010, from a native population in an area of abandoned pasture,
starting the regeneration of the forest, located 15 km from the capital Porto Velho, Rondonia.
The botanical identifications were made by botanist Dr. Jos Gomes from the herbarium of
the National Institute for Amazon Research, where a voucher specimen is stored under the
numbers 223344 for P. marginatum, 223303 for P. aleyreanum, 211724 for P. dilatatum,
221713 for P. aduncum and 211711 for P. alatabaccum. Essential oils (EO) were extracted
from fresh leaves by hydrodistillation for four hours in a Clevenger type apparatus. The oil

was dried over anhydrous sodium sulfate and, after filtration, stored at 4C until analysis.
2.2. Identification of essential oil components
The EO was analyzed by gas chromatography / mass spectrometry (GC/MS) to identify
their components. GC/MS was performed using a Hewlett-Packard 5971 instrument
employing C; the following conditions: column (30 m x 0,25 mm); carrier gas: He (1
mL/min); injector temperature: 250C; detector temperature: 200C; column temperature: 35-
180C at 4C/min then 180-250C at 10C/min; mass spectra: electron impact 70 eV.
Identification of the chemical components was based on the comparison of their mass spectral
with the National Institute for Standard Technology NIST62 library, comparison of
calculated retention indexes with literature values as described by Adams (2007).
2.3 Determination of anticandidal activity
Nine strains of Candida spp. were selected for testing (1 C. albicans, 3 C. tropicalis
and 5 C. parapsilosis), 3 of which were resistant to fluconazole. These strains are the most
commonly isolated from fungal infections in Cear State, Northeastern Brazil. The strains
were isolated from blood samples between 2009 and 2010 and belong to the Collection of
Yeasts of LABEL / FF / UFC (Laboratory of Bioprospection and Experiments in Yeast,
Faculty of Pharmacy, Federal University of Cear). Strains were inoculated on potato
agar (HiMed Mumbai-India) and incubated at 37 C/24 h. They were then plated on
CHROMagar Candida (HiMed Mumbai-India) to assess purity. The identification was made
by micromorphology on rice agar Tween 80, germ tube production, fermentation and
assimilation of carbohydrates as well as molecular tests. (Medrano et al., 2006; Menezes et
al., 2009)

Susceptibility tests of Candida spp. essential oils were performed according to protocol
M27-A3 by the method of microdilution in RPMI medium (pH 7.0) buffered with MOPS in
96-well plates. Each essential oil was weighed (160 mg) and dissolved in 4.0 mL of DMSO.
The solution was diluted in RPMI and the concentration of DMSO was not > 1%. The range
tested was from 10 to 0.018 mg/mL. The strains were exposed to EO and incubated at
37C/24 h. The MIC of EO was the lowest concentration that caused a reduction of 50% and
90% in of Candida spp. compared with the control. The strains C. parapsilosis ATCC 22019
and C. krusei ATCC 6258 were used as controls (CLSI, 2008a; CLSI, 2008b; Kiraz et al.,
2010). We used recently revised CLSI CBP values to identify strains of Candida resistant to
the fluconazole MIC results at > 4g/mL were defined as resistant for C. albicans, C.
parapsilosis, and C. tropicalis (Pfaller et al., 2010a,b; Pfaller et al., 2011).
2.4. Statistical analysis
The antifungal activity evaluated by broth microdilution method was expressed as mean
SD of MIC50 and MIC90, using a GraphPrism Program. The differences among the oils were
evaluated by Tukeys test with the significance level set at 5%. Retention indexes expressed
were calculated by linear regression using Kovats indexes of a hydrocarbon homologous
series, using the software Origin.
Results
Among the studied Piper species, a great diversity was found among the chemical
components. The structures of main compounds found in the oils are shown in Figure 1. For
the species P. aleyreanum, the major components were caryophyllene oxide (25.48%), E-
caryophyllene (15.11%) and limonene (6.28%). The species P. marginatum showed high

concentrations of 3,4-(methylenedioxy)propiophenone (27.08%), -copaene (23.98%) and E-
anethole (7.90%). For the species P. aduncum the major componentes were trans-isocrowecin
(32.02%), apiole (29.51%) and cis-isoelemicin (8.04%). The major componentes of P.
alatabaccum were methyl stearate (20.90%), methyl linoleate (17.90%), spathulenol (13.64%)
and E-caryophyllene (11.96%). For the species P. dilatatum the substances caryophyllene
oxide (22.62%), methyl stearate (19.79%) and methyl linoleate (18.31%) were the
components that prevailed as shown in Table 1.
Essential oils of Piper species from Amazon region showed the following MIC50
means: 0.37 0.1 mg/mL for P. dilatatum, which was the most active when evaluated by
Tukeys test, and 1.05 0.37 mg/mL for P. marginatum, 0.81 0.33 mg/mL for Piper
aleyreanum, 0.96 0.7 for P. aduncum and 0.90 0.45 for P. alatabacum. Besides P.
dilatatum, other essential oils did not show statistical differences in their antifungal activity
(Table 2).
3. Discussion
Candida spp. yeasts are usually isolated from biological samples and the amount of
available drugs is extremely limited. Pharmacological alternatives are needed to combat this
common infection. In addition to this, there are strains resistant to traditional antifungal
agents. Candida albicans is the major human and animal fungal pathogen and the effect of
antifungals essential oils from plants on this yeast is well known, many can be used in
conventional therapy, because the number of available antifungal agents is quite limited.
(Agarwal et al., 2010)
The epidemiology of systemic fungal infections (SFI) is changing, Candida albicans
yeast is still the most important, however, C. tropicalis, C. glabrata, C. parapsilosis and C.
krusei have been increasingly isolated and in some studies go beyond the isolation of C.

albicans. (Menezes, 2009; Diekma et al., 2009; Horn et al., 2009; Gomes et al., 2010;
Rodrguez et al., 2010; Sellami et al., 2011). In Cear the Candida spp. most commonly
isolated are C. albicans, C. tropicalis and C. parapsilosis (Menezes et al., 2009; Gomes et al.,
2010; Vasconcelos et al., 2011) were then chosen to perform the tests with essential oils
of Piper spp.
The main prescription antifungals for the treatment of IFS are fluconazole and
amphotericin (Horn et al., 2009). Candida spp. resistant to fluconazole have been detected in
various parts of the world (Pfaller et al., 2008; Pfaller et al., 2010b). Fluconazole is an
inexpensive drug with few side effects used in the treatment of infections caused by Candida
spp. The antifungal resistance has been found in several studies around the world, especially
resistance to fluconazole. In this study it was used strains of Candida spp. with elevated MICs
to fluconazole.
All oils showed activity against Candida spp. Essential oils are known to contain a
cocktail of natural monoterpenes, sesquiterpenes and hydrocarbons, with a variety of
functional groups that lead to an antifungal activity as described by Moreira (2004). The Piper
species are currently classified as major producers of essential oils (Silva et al, 2007).
The studied Piper species showed a great diversity of chemical components. Among
tested oils, the statistical comparison of anticandidal activities showed P. dilatatum as the
more active (Figure 2) with MIC50 in the range of 0.15-0.60 mg/mL as displayed in Table 2.
For P. dilatatum the main constituents are -copaene, cayophyllene oxide, methyl linoleate
and methyl stearate. Caryophyllene oxide, an oxygenated sesquiterpenoid, well known as
preservative in food, drugs and cosmetics, has been tested in vitro as an antifungal against
dermatophytes. Its antifungal activity has been compared to ciclopiroxolamine and
sulconazole, commonly used in onychomycosis treatment (Yang et al., 1999). Ten alkyl fatty
acid esters (AFAEs) were tested for in vivo antifungal activity against five plant diseases such

as rice blast, rice sheath blight, tomato gray mold, tomato late blight and barley powdery
mildew. Some AFAEs showed the most control efficacy against barley powdery mildew
among the tested plant diseases. Methyl and ethyl palmitates, methyl and ethyl oleates, methyl
linoleate, and methyl linolenate demonstrated both curative and protective activities against
barley powdery mildew (Choi et al., 2010). The essential oil of Piper diospyrifolium showed
as main constituent -copaene (47.73%), nevertheless the oil did not display antifungal
activity (Morandim et al., 2010). Taking into account the above information, caryophyllene
oxide and alkyl fatty acid esters probably respond for the antifungal activity of P. dilatatum.
The major constituents of P. aduncum oil were trans-isocrowecin, dill apiole and cis-
isoelemicin. In the study of other Piper species made by Morandim et al. (2010), P.
solmsianum showed the best activity and the major compound was E-isoelemicin (53.45%).
Rali et al. (2007) demonstrated the insecticidal activity of dill apoile against malaria mosquito
vector. Therefore, main compounds of P. aduncum essential oil, dill apiole and E-isoelemicin,
could be responsible for its antifungal activity.
The oil of P. marginatum showed MIC50 varying between 0.30-1.25 mg/mL. The
essential oil of P. marginatum showed as main constituents alpha-copaene and 3,4-
(methylenedioxy) propiophenone and (E)-anethole. The antifungal activity could be related to
(E)-anethole and minor compounds -pinene, -pinene E-ocimene and E-caryophyllene
(Huang et al., 2010; Moreira et al, 2007).
In conclusion, the Piperaceae family presents several species from Amazon forest used
in popular medicine in diseases caused by fungi, which use can be justified by the presence of
essential oils with antifungal activity and therefore could be pointed out as therapeutic
alternatives for the treatment of fungal infections caused by Candida spp.

Resumo
Neste trabalho foi avaliada a composio qumica e a atividade antifngica dos leos
essenciais de Piper marginatum, P. aleyreanum, P. dilatatum, P. aduncum e P. alatabaccum
oriundos da Amaznia contra cepas de Candida tropicalis isoladas de aves domsticas
brasileiras. O projeto foi submetido e aprovado pelo Comit de tica para o Uso de Animais
da Universidade Estadual do Cear. A anlise qumica dos leos foi feita por cromatografia
gasosa acoplada espectrometria de massa (CG/EM). Foram utilizadas 16 aves entre elas 04
marrecos, 01 galo e 11 galinhas. Foi relizada coleta de secreo da microbiota oral das aves
com utilizao de swab descartvel. Os leos essenciais foram testados na faixa de 10 a 0,015
mg/ml pelo mtodo de microdiluio em caldo de acordo com o protocolo M27-A3. O MIC
do OE foi a menor concentrao que causou uma reduo de 50% e 90% de C. tropicalis
comparado com o controle. Para a espcie P. aleyreanum, os componentes principais foram
xido de cariofileno (25,48%), E-cariofileno (15,11%) e limoneno (6,28%). A espcie P.
marginatum mostrou altas concentraes de metil eugenol (27,08%), -copaeno (23,98%) e
E-anetol (7,90%). Para a espcie P. aduncum os componentes principais foram trans-
isocrowecin (32,02%), apiole (29,51%) e cis-isoelemicin (8,04%). Os componentes principais
para P. alatabaccum foram metil octadecanoate (20,90%), metil linoleato (17,90%),
spathulenol (13,64%) e E-cariofileno (11,96%). Para a espcie P. dilatatum prevaleceram as
substncias xido de cariofileno (22,62%), metil-octadecanoate (19,79%) e metil linoleato
(18,31%). De acordo com o clculo das mdias geomtricas para atividade com MIC50 os
leos essenciais de espcies de Piper da Amaznia mostraram os seguintes resultados: P.
dilatatum que foi o mais ativo a MG foi 278, para as espcies P. aleyreanum, P. marginatum,
P. aduncum e P. alatabaccum as GMs foram respectivamente 703, 1340, 786 e 650. Todos
os leos mostraram atividade contra Candida tropicalis.

Abstract
The increasing incidence of infections caused by fungi in domestic birds, along with the
resistance that these antimicrobial agents have developed, has led to constant search for
effective alternative treatment that may offer better treatment option. The essential oil of five
Piper species grown in the Amazon forest were extracted and tested against Candida
tropicalis the main fungus present in poultry oral microbiota. The essential oils of Piper
species showed a great diversity of chemical components such as sesquiterpenes, aromatic
compounds and fatty acid methyl esters. Among tested oils Piper dilatatum showed the best
MIC50 with activity in the range of 18-600 g/ml. The main constituents of P. dilatatum are -
copaene, cayophyllene oxide, methyl linoleate and methyl octadecanoate. Piperaceae Family
contains several species rich in essential oils with antifungal potential, which could be
therapeutic alternatives for the treatment of fungal infections that attack domestic birds caused
by C. tropicalis.

Introduction
Candida tropicalis is one of the predominant species between the not-albicans related to
candidemia (Alfonso et al., 2010) and it is usually found in the gastrointestinal tract of birds,
being one of the species most commonly isolated in these animals, however the population
imbalance of these yeasts causes problems in the digestive tract, and may also infect the
reproductive tract and eye, as well as become systemic (Shrubsole-Cockwill, 2010). Rare and
serious cases can cause deformities in beak causing the bird to not use it appropriately
difficulting or even preventing normal feeding (Osrio, 2007; Godoy, 2001).

effective treatment alternatives that may offer better treatment options (Bautista-Baos et al.,
2003).
The Amazon region of Brazil presents a great plant genetic potential, therefore, a
relevant value for the discovery of substances with medicinal activity (Azevedo et al., 2006;
Lucena et al., 2007; Teklehaymanot et al., 2007).
Piperaceae Family which comprises 12 genres and 1400 species with mainly a
pantropical distribution stands out among the bioproduction which come from Amazon. The
genre Piper is the largest of the family, with 700 species. Between them, there are 170 species
that occur in Brazil. The Piperaceae is represented by herbs, lianas, shrubs, and rarely by trees
(Andrade et al., 2008).
Piper amalago leaves are used in Mexico and in Brazil to relieve stomach pains and
fight various infections. In the State of Paraba, leaves and stalks of Piper marginatum and
Piper tuberculatum are used against snake bite and as a sedative (Guimares et al., 2004).
Chemical studies showed that the Piper genus has many components, including flavonoids,
unsaturated amides, lignans, aristolactams, long and short chain esters, terpenes, steroids and

alkaloids (Facundo et al., 2005). Terpenes and phenolic compounds in general are responsible
for the antimicrobial activity demonstrated in vegetable oils. The antimicrobial activity of
essential oils in vitro, both and in vivo, has justified the research on traditional medicine
focused on the characterization of its antimicrobial activity (Agarwal et al., 2010).
This work evaluated the antifungal activity of essential oils of P. marginatum Jacq, P.
aleyreanum C. DC., P. dilatatum L. C. Rich., P. aduncum L. and P. alatabaccum Trel &
Yuncker against Candida tropicalis strains obtained from domestic birds.
Materials and methods
Plant materials and isolation of essential oil
The leaves of the species P. marginatum, P. aleyreanum, P. dilatatum, P. aduncum and,
P. alatabaccum were collected from the native population in an area of abandoned pasture,
where started the regeneration of the forest, located 15 km from the capital, Porto Velho,
Rondonia. The botanical identifications were made in the herbarium of the National Institute
for Amazon Research, where the voucher specimen is stored under the numbers 223344 for P.
marginatum, 223303 for P. aleyreanum, 211724 for P. dilatatum, 221713 for P. aduncum and
211711 for P. alatabaccum. Essential oils (EO) from fresh leaves were extracted by
hydrodistillation for four hours in a Clevenger type apparatus (Craveiro et al., 1976). The oil
was dried over anhydrous sodium sulfate and, after filtration, stored at 4 C until and analyzed.
Identification of oil components
The oils were analyzed by gas cromatography-mass spectrometry (GC/MS) to identify
their components. GC/MS was performed using a Hewlett-Packard 5971 instrument
employing C; the following conditions: column (30 m x 0.25 mm); carrier gas: He (1 ml/min);
injector temperature: 250 C; detector temperature: 200c; column temperature: 35-180 C at

4 C/min then 180-250 C at 10 C/min; mass spectra: electron impact 70 eV. Identification
of the chemical components was based on the comparison of their mass spectral with the
National Institute for Standard Technology NIST62 library and comparison of calculated
retention indexes with literature values (Adams, 2007).
Fungal strains
16 birds were used including teal (4), cock (1) and hen (11). The project was submitted
to the Ethics Committee for the use of domestic animals of the University of Cear State
under number 11044867-7/29 and approved in August 25, 2011. Collection of
microorganisms of oral microbiota secretion collection of birds was carried out with use of
disposable swab. The material collected was sown in potato agar (Himedia Mumbai-India) in
test tubes previously prepared. Then they were sown on CHROMagar Candida (Himedia
Mumbai-India) to assess purity. The identification was done via commercial system
Auxacolor and confirmed by rice agar micromorphology (Medrano et al.; 2006; Menezes et
al.; 2009).
Anticandidal activity
C. tropicalis strains, isolated from poultry oral microbiota, were selected for testing.
The strains were inoculated in potato dextrose agar (Himedia Mumbai-India) and incubated at
37oC /24 h and sensitivity testing of strains of C. tropicalis essential oils was carried out in
accordance with the M27-A3 Protocol, by the method of microdiluition in RPMI broth (pH
7.0), buffered with MOPS in 96-well plates. Each EO (160 mg) was weighed and dissolved in
DMSO (1.0 ml). The solution was diluted in RPMI medium and DMSO concentration was
not higher than 1%. The range tested varied from 10.000 to 18 g/ml. The strains were
exposed to EO and incubated at 35C /24 h. The MIC of the EO was the lowest concentration

that caused a 50% reduction of Candida growth spp. when compared with the control. The
strains C. parapsilosis ATCC 22019 and C. krusei ATCC 6258 were used as controls (CLSI,
2008a; CLSI, 2008b; Kiraz et al.; 2010).
Results
The studied Piper species showed a great diversity among chemical components. The
essential oil composition of these species was displayed in Table 1. For the species P.
aleyreanum, the major components were caryophyllene oxide (25.48%), caryophyllene
(15.11%) and-limonene (6.28%). The species P. marginatum showed high concentrations of
methyl eugenol (27.08%) -copaene (23.98%) and E-anethole (7.90%). For the species P.
aduncum the major components were trans-isocrowecin (32.02%), apiole (29.51%) and cis-
isoelemicin (8.04%). The major components of P. alatabaccum were methyl octadecanoate
(20.90%), methyl linoleate (17.90%), spathulenol (13.64%) and-caryophyllene (11.96%). For
the species P. dilatatum the substances caryophyllene oxide (22.62%), methyl octadecanoate
(19.79%)-and methyl linoleate (18.31%) were the components that prevailed.
Piper essential oils of species from the Amazon region showed the following MIC50
means: 278 g/ml for P. dilatatum, the most active when evaluated by Tukey's test, and 1340
g/ml for P. marginatum, 703 g/ml for P. aleyreanum, 786 g/ml for P. aduncum and 650
g/ml for P. alatabacum. Besides P. dilatatum, other essential oils did not show statistical
differences in their antifungal activity.
Discussion
In animals as in humans, mycosis is an opportunistic infection frequently becoming a
clinical problem despite improvements in veterinary medical technology and prevention.
Deep mycoses such as candidiasis, have been reported in farm animals such as cows, horses

and pigs and pets such as birds, dogs and cats. The diagnosis of these diseases is often based
on clinical signs, and on abnormal histological and mycological findings. Due to contact with
people, animals may represent a source of fungal infections in human being therefore
alternatives for the control of these infections are relevant to this public health problem
(Ishibashi et al., 2010).
Candida spp. yeasts are usually isolated from biological samples and the amount of
available drugs is extremely limited. Pharmacological alternatives are needed to combat this
common infection. In addition to this, there are strains resistant to traditional antifungal
agents. The effect of antifungal essential oils from plants on this yeast is well known, and
many have been used in conventional therapy (Agarwal et al., 2010).
Candida spp. is part of the normal intestinal microbiota of healthy birds and is typically
found in the upper gastrointestinal tract, primarily the oropharynx, crop, and esophagus
(Samour et al., 2002; Velasco, 2000). C. tropicalis is an opportunistic diploid fungus, with a
worldwide distribution, and is one the primary cause of candidiasis, which is often associated
with immunosuppression. It is often isolated from poultry droppings; however the lack of
specific care with cleaning breeding enables fungal species get contact with the oral cavity of
birds (Shrubsole-Cockwill, 2010; Hublek, 2004). In this study we evaluated the antifungal
activity of Piper spp. against strains of C. tropicalis from domestic birds.
